Note:本发明公开了一种过表达海栖热袍菌乙酰乳酸合酶催化亚基的菌株的构建方法及其用途。重组菌株的构建方法包括以下步骤:根据TmAHAS-CSU的基因序列设计上下游引物,设计的上游引物带有NdeI酶切位点,下游引物带有BamHⅠ酶切位点,然后以海栖热袍菌基因组DNA为模板,通过PCR扩增技术得到目的基因片断;将目的片断连接到表达载体pET28a上,得到重组质粒pET28a-TmAHAS-CSU,并通过热击法将该重组质粒转化入感受态细胞中以构建过表达Tm-AHAS-CSU蛋白的重组菌株。在此基础上验证了该重组菌株的功能及它在立体选择性地催化合成R-PAC及其类似物方面的用途。
